To help minimize non-specific product formation (including primer-dimers), our SYBR® Green amplification reagents use the hot-start enzyme Taq DNA polymerases. Step 4: Design Primers
An assay contains 2 primers (for SYBR chemistry) or 2 primers and 1 TaqMan® probe (for TaqMan® chemistry) and it's used to amplify and detect the specific target during the PCR reaction by emission of a fluorescent signal.

The Primer Express® Software v3.0 allows you to design your own primers and probes using TaqMan® and SYBR® Green I dye chemistries for gene quantitation and allelic discrimination (SNP) real-time PCR applications. Developed specifically for use with our 7300, 7500, 7500 Fast, and 7900HT Fast Real-Time PCR Systems, Primer Express® Software provides customized application-specific documents for absolute/relative quantitation and allelic discrimination. This latest software version includes an improved workflow for allelic discrimination assay designs and supports the Windows XP® operating system.
Step 5: Run & Analyze
The PCR reaction is performed on a real-time PCR instrument and the fluorescent signal generated during the PCR reaction is analyzed by the software to determine a Ct for each sample (Ct = cycle threshold).

Instrument Calibration Kits

Optimal performance from your Applied Biosystems Real Time PCR system requires calibration upon installation and periodically there after. Real Time PCR systems must be calibrated for region of interest (ROI), background fluorescence, optical quality, and dye configuration.
